Background
IFT88 (intraflagellar transport protein 88) is an 833-amino-acid tetratricopeptide-repeat protein that serves as a core subunit of the IFT-B complex, powering anterograde delivery of ciliary cargo by kinesin motors, and is therefore indispensable for assembly and maintenance of motile and primary cilia; beyond this canonical ciliary role, IFT88 also localizes to centrosomes in proliferating non-ciliated cells where it binds the Rb regulator Che-1 to restrain G1-S transition, acts in mitotic spindle orientation, endocytic recycling of immune receptors, angiogenesis, and cartilage homeostasis, while mutations or depletion of IFT88 produce developmental malformations, polycystic kidney disease, skeletal defects, retinal degeneration, and tumor progression, illustrating its broad importance in both cilium-dependent and cilium-independent cellular processes.
